In order to determine the epitopes of Fel d 1 recognized by REGN1908 and REGN1909, HDX studies were performed for each antibody co-complexed with rFel d 1.mmh. The solution was then quenched in cold acidic aqueous solution to minimize back-exchange and subjected to proteolysis and mass spectrometry analysis. Fel d 1-derived peptic peptides that exhibited increased mass greater than 0.2 mass units relative to control experiment were defined as the binding epitopes based on H/DX methodology.

The sole purpose of an immune system is to protect itself by killing any bacteria, fungi, virus or any other seriously dangerous, infectious microorganisms, that try to get into our bodies. When people are allergic to a certain substance, such as pollen, the immune system starts attacking the foreign microorganism, by making antibodies that kill the substance. Due to this, our bodies react with having mild to serious symptoms like a runny noses, itchy eyes or wheezing, which can cause the body to develop asthma later on in life. Food allergies are one of the most common allergies.

Allergic conditions seem to be becoming more common in the UK. Whats behind the rise, and how can pharmacists help this growing number of patients?

Professor Sheena Cruickshank, academic lead for public engagement at the British Society of Immunology, says developed countries are seeing an increase in autoimmune diseases such as allergies, which a 2010 study in the Clinical and Experimental Immunology journal supports. The big question is why.

In an autoimmune disease such as an allergy, the immune system reacts to a benign trigger it should ignore , resulting in inflammation. This causes symptoms ranging from swelling to mucus production, redness and soreness. In severe cases, allergic reactions can lead to anaphylactic shock, which can be fatal. Common allergies include hayfever, eczema, contact dermatitis, asthma and food allergies.

An allergy is not the same as intolerance, which patients may have, to certain foods for example, where someone may lack an enzyme required to digest a protein, such as lactose.

Is the hygiene hypothesis outdated?

However, scientists now view the hygiene hypothesis as outdated.

According to Hedberg, an estimated 35 percent of all women and 17 percent of all men currently have developed a nickel allergy. Nickel allergy is the commonest form of contact allergy. If you have it, you can get dermatitis from contact with the metal in objects such as glasses, watches, belts, tools and keys. And we must not forget that a further 4,300 chemicals in our society can trigger contact allergies A contact allergy always arises where the substance has been in contact with the skin, but often only a couple of days or even weeks later. This means it can take a bit of detective work to work out what has caused the allergy. We need to raise public awareness of what a delayed contact allergy actually means. Many people think they have suffered a direct allergy when they get a skin rash, but in the case of contact allergies, the reaction can be delayed several weeks, says Hedberg.My best advice is always to perform allergy tests as recommended on the packaging before any beauty treatment such as hair colouring, piercing, false nails, false eyelashes, face masks or getting tattoos. You also need to wait at least a couple of days to see whether you have a possible allergy. Serious allergic reactions can be life threatening in the worst case.

House Dust Mite Minimisation

The first room to tackle is the bedroom and in particular the bedding, where we spend the greatest number of consecutive hours. A combination of the following four measures is recommended:

  • Wash sheets, pillow cases and other bedding weekly in hot water . This will kill dust mites and wash away the allergen they produce. If you cannot wash in hot water, use a commercial product containing tea tree or eucalyptus oils, formulated to kill dust mites in cold water. If washing normally, hot tumble drying of washed items for ten minutes after they are dry, will kill dust mites. Dry cleaning is not as effective as it will kill house dust mites but won’t remove the allergen they produce.
  • Cover mattress, pillow and quilt with dust mite resistant covers. Some health funds may provide a rebate for the purchase of these items. The covers must be washed every two months. If covers are not available, wash blankets and non-encased washable doonas every three months in hot water.
  • Remove sheepskins or woolen underlays from the bed and bedroom.
  • Remove all soft toys from the bed and bedroom. Replace them with wooden or plastic toys which can be washed. If keeping soft toys, wash them in eucalyptus oil weekly or place in the freezer overnight). Freezing soft toys overnight kills mites but does not remove the allergen.

There was great excitement in the laboratory when researchers from Aarhus University recently discovered the unique mechanisms of an antibody that blocks the immune effect behind allergic reactions.

The team of researchers from the Departments of Engineering and Molecular Biology and Genetics together with German researchers from Marburg/Giessen has now described the molecular structure and mechanisms of action of the antibody, and the results are surprising.

They were hoping to find new methods to improve existing treatment, but instead they identified how a specific antibody is apparently able to completely inactivate the allergic processes.

The antibody interacts in a complex biochemical process in the human body by which it prevents the human allergy antibody from attaching to cells, thus keeping all allergic symptoms from occurring.

Allergic effects of birch pollen and insect venom eliminated

Furthermore, the researchers have observed that the antibody also removes the IgE molecules even after binding to its receptors.
